ABSTRACT

Background & objectives: Bacterial vaginosis (BV) involves the presence of a thick vaginal multispecies biofilm, where Gardnerella vaginalis is the predominant species. The reason for an increase in the number of G. vaginalis which are usually present as normal flora of the female genital tract in cases of BV, is not known. Hence, the objective of the present study was to compare the biotypes and virulence factors of G. vaginalis isolated from the genital tract of women with and without BV. Methods: High vaginal swabs collected from 811 women of reproductive age were cultured. G. vaginalis isolates were biotyped and tested for adherence to vaginal epithelial cells, biofilm formation, agglutination of human red blood cells (RBCs), protease production, phospholipase production and surface hydrophobicity. Results: Of the isolates from women with BV, 83.3 per cent (60/72) showed good adherence, 78.4 per cent (58/74) produced biofilm, 82.9 per cent (63/76) produced phospholipase, 67.1 per cent (51/76) produced protease, 77.3 per cent (58/75) were positive for surface hydrophobicity and 61.6 per cent (45/73) were positive for haemagglutination of human RBC. In case of G. vaginalis from non-BV women, 25 per cent (15/60) isolates showed good adherence, 18.4 per cent (9/49) biofilm production, 35 per cent (21/60) phospholipase, 36.6 per cent (22/60) protease, 41.7 per cent (25/60) surface hydrophobicity and 10.1 per cent (6/59) agglutination of human RBCs. Maximum number of isolates belonged to biotypes 6, 2 and 3. Biotype 3 was more associated with non-BV rather than BV; biotype 6, 2 and 1 were more associated with cases of BV. Maximum virulence factors were expressed by biotypes 6, 2 and 1. Interpretation & conclusions: Virulence factors were more expressed by G. vaginalis isolates obtained from women with BV rather than from non-BV. Biotypes 6, 2 and 1 were more associated with cases of BV and expressed maximum virulence factors.

ABSTRACT

There is a growing demand for invasive procedures involving the inferior vena cava, in particular for placement of vena cava filters. It is not always easy to identify the more distal renal vein with cavography, for safe release of filters. OBJECTIVES: To determine parameters for the relationships between the renal veins and the infrarenal vena cava and their corresponding vertebral bodies, their relationships with biotype and the occurrence of anatomic variations, the relationships between vertebral bodies and the bifurcation of the common iliac veins and the distance from this bifurcation to the outflow of the more distal renal vein, with reference to placement of vena cava filters. METHODS: A total of 150 abdominal computed tomography scans conducted from October to November 2011 were analyzed and classified according to the biotype exhibited (using Charpy's angle). Scans were performed at MEDIMAGEM and analyzed at the Integrated Vascular Surgery Service, both part of Hospital da Beneficência Portuguesa in São Paulo, Brazil. RESULTS: In 127 of the 150 scans analyzed (84.66%), the more distal renal vein emerged between the first lumbar intervertebral space (L1-L2) and the body of L2, irrespective of patient biotype. Just 23 patients (15.33%) exhibited a more distal renal vein with outflow below the body of L2, i.e. in the projection of the space between L2 and L3. CONCLUSIONS: The radiological correlation between the confluence of the more distal renal vein and vertebral bodies exhibits little variation, irrespective of the biotype of the patient...


Há uma demanda crescente por procedimentos invasivos que abordam a veia cava inferior, especialmente o implante de filtros de veia cava. A identificação da veia renal mais caudal para a liberação segura do filtro nem sempre é fácil durante a cavografia. OBJETIVOS: Estabelecer parâmetros da relação das veias renais e da cava infrarrenal com o corpo vertebral correspondente, sua relação com a biotipologia, presença de variações anatômicas, relação dos corpos vertebrais com a bifurcação das veias ilíacas comuns para a veia cava e distância desta bifurcação até a desembocadura da veia renal mais caudal, visando à implantação de filtro de veia cava. MÉTODOS: Foram analisadas 150 tomografias computadorizadas de abdome no período entre outubro e novembro de 2011, tendo sido agrupadas de acordo com o biotipo apresentado (ângulo de Charpy). As tomografias forem realizadas na MEDIMAGEM e analisadas no Serviço de Cirurgia Vascular Integrada, ambas da Beneficência Portuguesa de São Paulo. RESULTADOS: Dos 150 exames analisados, 127 (84,66%) apresentaram a emergência da veia renal mais caudal desde a projeção do primeiro espaço intervertebral lombar (L1-L2) até o corpo de L2, independentemente do biotipo do paciente. Somente 23 pacientes (15,33%) apresentaram a desembocadura da veia renal mais caudal abaixo do corpo de L2, ou seja, na projeção do espaço entre L2 e L3. CONCLUSÕES: A correlação radiológica da confluência da veia renal mais distal em relação aos corpos vertebrais apresenta pouca variação, independentemente do biotipo do paciente...

ABSTRACT

La introducción de la vacuna contra Haemophilus influenzae tipo b en los programas de inmunización de muchos países produjo una reducción marcada en la incidencia de enfermedad invasiva causada por este serotipo y en su portación y un incremento de otros tipos capsulares y de aislamientos no capsulados. Se estudiaron 313 aislamientos de H. influenzae recuperados de sitio estéril, provenientes de pacientes pediátricos y adultos con enfermedad invasiva atendidos en 90 hospitales de la Red Nacional de Laboratorios para Meningitis e Infecciones Respiratorias Agudas Bacterianas durante el período 2005-2010. Las patologías más frecuentes fueron neumonía, 40,3 % (n = 126), meningitis, 30,0 % (n = 94) y bacteriemia, 26,5 % (n = 83). En los pacientes pediátricos (n = 279), la mayor frecuencia de aislamientos correspondió a menores de 2 años, 74,5 % (n = 208). Con respecto a la distribución de tipos, el 61,3 %, correspondió a H. influenzae no capsulados (n = 192); el 20,1 % al b (n = 63); 11,2 % al a (n = 35); 4,8 % al f y 2,6 % a otros. En meningitis predominaron H. influenzae capsulados mientras que en neumonía y bacteriemia resultaron dominantes los tipos no capsulados. Se determinó el biotipo en 306 aislamientos. Todos los aislamientos de tipo a correspondieron al biotipo II; el 66,7 % de los tipo b pertenecieron al biotipo I. Mediante las técnicas de aglutinación en lámina y PCR se estudiaron 220 aislamientos; la concordancia entre ambas fue de 0,982 (IC: 0,92-1,00). En el último año se encontró un aumento significativo del tipo b, lo cual indica la importancia de mantener la vigilancia clínica y laboratorial de la enfermedad invasiva por H. influenzae.


The introduction of the Haemophilus influenzae type b vaccine in the immunization programs of many countries has greatly reduced this invasive disease and the carriage caused by this serotype, also increasing other capsular types and non-capsular isolations. There were 313 isolations of H. influenzae under study, which were recovered from a sterile site coming from pediatric and adult patients carrying the invasive disease. Patients were treated at 90 different hospitals belonging to the Red Nacional de Laboratorios para Meningitis e Infecciones Respiratorias Agudas Bacterianas (National Lab Network for Meningitis and Acute Bacterial Respiratory Infections) from 2005 to 2010 for the following disorders: pneumonia, 40.3% (n = 126), meningitis, 30.0% (n = 94) and bacteremia, 26.5% (n = 83). In pediatric patients (n = 279), the highest frequency of isolations corresponded to children under the age of 2 years, 74.5% (n = 208). Regarding type distribution, 61.3% corresponded to non-capsular H. influenzae (n = 192), 20.1% to type b (n = 63), 11.2% to type a (n = 35), 4.8% to type f, and 2.6% to other types. Capsular H. influenzae was predominant in meningitis whereas non-capsular H. influenzae in pneumonia and bacteremia. The biotype was determined in 306 isolations. The totality (100%) of type a (n = 35) was biotype II whereas 66.7% of type b (n = 63) was biotype I. Slide agglutination and PCR tests were used in 220 isolations. There was a match of 0.982 (IC: 0.92-1.00) between them. During the last year, there was a great increase in type b, showing the importance of clinical and laboratory-based surveillance of the invasive disease caused by H. influenzae.

Background & objectives: El Tor Vibrio cholerae O1 carrying ctxBC trait, so-called El Tor variant that causes more severe symptoms than the prototype El Tor strain, first detected in Bangladesh was later shown to have emerged in India in 1992. Subsequently, similar V. cholerae strains were isolated in other countries in Asia and Africa. Thus, it was of interest to investigate the characteristics of V. cholerae O1 strains isolated chronologically (from 1986 to 2009) in Thailand. Methods: A total of 330 V. cholerae O1 Thailand strains from hospitalized patients with cholera isolated during 1986 to 2009 were subjected to conventional biotyping i.e., susceptibility to polymyxin B, chicken erythrocyte agglutination (CCA) and Voges-Proskauer (VP) test. The presence of ctxA, ctxB, zot, ace, toxR, tcpAC, tcpAE, hlyAC and hlyAE were examined by PCR. Mismatch amplification mutation assay (MAMA) - and conventional- PCRs were used for differentiating ctxB and rstR alleles. Results: All 330 strains carried the El Tor virulence gene signature. Among these, 266 strains were typical El Tor (resistant to 50 units of polymyxin B and positive for CCA and VP test) while 64 had mixed classical and El Tor phenotypes (hybrid biotype). Combined MAMA-PCR and the conventional biotyping methods revealed that 36 strains of 1986-1992 were either typical El Tor, hybrid, El Tor variant or unclassified biotype. The hybrid strains were present during 1986-2004. El Tor variant strains were found in 1992, the same year when the typical El Tor strains disappeared. All 294 strains of 1993-2009 carried ctxBC ; 237 were El Tor variant and 57 were hybrid. Interpretation & conclusions: In Thailand, hybrid V. cholerae O1 (mixed biotypes), was found since 1986. Circulating strains, however, are predominantly El Tor variant (El Tor biotype with ctxBC).

ABSTRACT

The present study was conducted to correlate the biotypes of Gardnerella vaginalis strains isolated from cases of bacterial vaginosis and their virulence factors. Thirty-two strains of G. vaginalis isolated from cases of bacterial vaginosis were biotyped. Adherence to vaginal epithelial cells, biofilm production, surface hydrophobicity, phospholipase C and protease activity were tested on these isolates. Biotype 1 was the most prevalent (8; 25%), followed by biotype 2 (7; 21.9%) and biotypes 5 and 8 (5; 15.6%). We did not find any statistical correlation between G. vaginalis biotypes and its virulence factors. Virulence factors expressed by G. vaginalis were not associated with a single biotype.

ABSTRACT

El lector del panel microbiológico automatizado autoScan®-4 detecta el crecimiento del hongo en diversos sustratos bioquímicos, presentes en los pozos de los paneles del MicroScan®. El propósito de este estudio fue relacionar el ¨biotipo¨ identificado por el MicroScan® con la especie causante de criptococosis, con el objeto de permitir una identificación en el menor tiempo posible. Se evaluaron 82 cepas de Cryptococcus spp. aisladas a partir de muestras clínicas entre 1995 y 2004. Para garantizar la pureza de las cepas, se realizó la identificación de las mismas por métodos convencionales; para identificar las especies se utilizó el medio L-canavanina-glicina-azul de bromotimol (CGB). Se utilizó también el panel de identificación rápida de levaduras del MicroScan®, con el fin de determinar el ¨biotipo¨. El MicroScan® reveló 27 diferentes ¨biotipos¨. De los 82 aislados tipificados con el uso del medio CGB, el 91,46 % correspondieron a C. neoformans y 8,54 % a C. gattii. No se encontró una diferencia significativa entre los ¨biotipos¨ y las especies (p>0,05). Sin embargo, se encontró significancia estadística entre las especies C. gattii y C. neoformans y la asimilación de p-nitrofenil-N-acetil-ß-D-glucosamina (NAG) (p<0,05). El panel de identificación rápida de levaduras del MicroScan® no fue capaz de diferenciar ambas especies.


The automated reader of the AutoScan® -4 microbiological panels detects fungi growth in various biological substrates present in the MicroScan® panel wells. The purpose of this study was to relate the biotype identified by the MicroScan® with the species causing the criptococcosis, in order to obtain identification in the shortest possible period. The evaluation included 82 Cryptococcus spp. strains isolated from clinical samples between 1995 and 2004. To guarantee the purity of the strains they were also identified by conventional methods; to identify the species we used L-canavanin-glycine-bromthymol blue medium (CGB). We also used the fast yeast identification MicroScan® panel with the purpose of determining the “biotype”. The MicroScan® revealed 27 different “biotypes”. Of the 82 isolates typed with the CGB medium, 91.46% corresponded to C. neoformans and 8.5% to C. gattii. No significant difference was found among the “biotypes” and the species (p<0,05). Nevertheless, there was a statistically significant difference between the assimilation of p-nitrophenil-N-acetyl-ß-D-glucosamine (NAG) (p<0.05) by the C. gattii and the C. neoformans species. The fast yeast identification MicroScan® panel was not able to differentiate between both species.

BACKGROUND: The association of Streptococcus bovis biotypes with the type of clinical infection and underlying malignancies and data on antimicrobial susceptibility of S. bovis have rarely been reported in Korea. The aim of this investigation was to characterize the clinical features of patients with S. bovis bacteremia, and to determine the antimicrobial susceptibility of S. bovis strains isolated from blood cultures. METHODS: The clinical data of 67 S. bovis isolates between May 1998 and April 2005 at Wonju Christian Hospital were retrospectively analyzed. The organism was identified by API Strep 32 kit and, for blood isolates, antimicrobial susceptibility testing was performed by the disk diffusion method and penicillin MICs were determined by E test. RESULTS: Of the 67 S. bovis isolates, 18 (27%) were biotype I and 49 (73%) were biotype II. Isolation rates by specimen type were, in decreasing order, wound. 37%; blood, 19%; and urine, 12%. Of the 13 S. bovis bacteremias, 2 were caused by biotype I and 11 were by biotype II; liver diseases (46%) were the most common underlying diseases; none of the 13 patients had gastrointestinal malignancies; one and three isolates were intermediate and resistant to penicillin, respectively; eleven were resistant to erythromycin; two and five were intermediate and resistant to clindamycin, respectively. CONCLUSION: Most of the S. bovis isolates from blood were biotype II. Liver diseases were the most common underlying diseases. S. bovis isolates from blood displayed a high rate of resistance to erythromycin and clindamycin.

ABSTRACT

BACKGROUND: Emerging drug resistance and increasing incidence of enterococci infection necessitates an accurate species identification. METHOD: We compared the identification of twelve strains of American Type Culture Coliection(ATCC) (E. faecalis, E. avium, E. faecium, E. raffinosus, E. durans, E. casseliflavus, E. hirae, E. flavescens, E. malodoratus, E. gallnarum, E. mundtii, and E. sulfureus) and 73 clinical isolates of enterococci(E. faecalis 8, E. faecium 8, E. avium 13, E. durans 5, and unidentified strains 39) by Vitek Gram-positive Identification card (version R08.1, bio- Merieux Vitek, Hazelwood, Mo., U.S.A.), ribotyping and conventional scheme of Facklam and Sahm. RESULTS: All ATCC strains could be identified on the basis of characteristic 16S-23S rDNA fingerprint patterns by using Eel I and Hind III. Of 39 strains unidentified by Vitek GPI card, 25 strains were identified as E. faecium, 5 strains were E. gallinarum, 5 strains were E. casseliflavus, 3 strains were E. avium and 1 strain was E. hirae. CONCLUSION: By ribotyping with Eel I and Hind m digestion, all twelve ATCC strains and 73 clinical strains were identified and it seems to be a reliable method for identification of Enterococcus species.

ABSTRACT

Foram analisadas as características de 386 cepas de Corynebacterium diphtheriae isoladas na Seção de Bacteriologia do Instituto Adolfo Lutz, São Paulo, no período de 1980 a 1986. Verificou-se que 58,3% destas cepas foram capazes de fermentar a sacarose. O biótipo mais frequente foi o mitis (52,2%), seguidos pelos biótipos intermedius (26,4%), gravis (9,9%) e de cepas de comportamento atípico (11,7%). Das cepas pertencentes ao biótipo intermedsus, 75,5% foram capazes de fermentar a sacarose. Com relação à produção de toxina, detectada pelo método de Elek, verificou-se que 92,8% das cepas foram poligênicas e que 97,3% das cepas fermentadoras de sacarose produziram toxina (AU).

Foram isoladas de material humano 1.000 cepas de Salmonella typhimurium, durante o período de 13 meses, começando em agosto de 1976. Dentre as cepas isoladas, 89 (8,90/0) não descarboxilaram a lisina. Sugere-se que as cepas lisina descarboxilase negativas estariam associadas a infecções hospitalares (AU).
